Actiunea metotrexatului asupra metabolismului celular

THE METOTHREXAT ACTION ON CELL METABOLISM. In this study we have followed the level of reactive oxygen species to patients under cytotoxic therapy with metothrexat (MTX). Mitochondria is the major intracellular source of reactive oxygen species (ROS) and free radicals due to electron leakage from the respiratory chain. In the normal physiological conditions, a small fraction of the oxygen consumed by mitochondria is constantly converted to superoxid anions, hydrogen superoxide, hydroxil radicals, and other ROS. An excess production of ROS is harmful to cells, becouse it causes the impairment of electron transport chain and the production of adenosin triphosphate (ATP). MDA (malondialdehide) determination in blood serum, is a standard method to estimate the oxidative stress and the level of reactive oxygen species. Mitochondrial defects have long been suspected to play an important role in the development and progression of cancer. At the examined patients, for malign affections, the increasing of the reactive oxygen species appeared after cytotoxic therapy, but the MTX metabolism was normal.
